I imagine that you want the following to evaluate

Conjugate[a + I*b]

Conjugate[a + I*b]

This would require that a and b are real.  If they are, there are a
couple of  approaches:

altConjugate[x_] := 
  x /. Complex[re_, im_] :> Complex[re, -im]

{altConjugate[I], altConjugate[3 + 4*I], 
  altConjugate[a + b*I]}

{-I, 3 - 4*I, a - I*b}

or

altConjugate[x_] := ComplexExpand[Conjugate[x]]

{altConjugate[I], altConjugate[3 + 4*I], 
  altConjugate[a + b*I]}

{-I, 3 - 4*I, a - I*b}
