Here is one way. Matrix of operators:

  operator = {{D[#, x]& , 0 # & }, {D[#, y]& , D[#, x]& }}

Matrix of functions:

  functions = {{x y, x}, {x + y, 3}}

Compose them:

  Inner[Compose, operator, functions]
