Hi,
Mathematica 5.2 supports this Automatically.
If you have multicore system while installing Mathematica it will install the files needed to support this, you need to know anything else as a user, nor to program differently in Mathematica, since implementation of internal functions will take use of it.
However, if you have an access multiple processor machines you can use the Parallel Computing ToolLit
