Dear Sam,

Try:

Assuming[w/k < KK && k > 0 && KK > 0 && w > 0,  Simplify[-k^2 + w^2/KK^2 > 0]]

or

Simplify[-k^2 + w^2/KK^2 > 0, w/k < KK && w > 0 && k > 0 && KK > 0]

Simplify needs to be inside Assuming for evaluation.
