I am not sure what you really want to do, but here is one way to
generate a new set of vertices everytime the plot command is executed:

myvertices := Map[Point[#] &, Table[{Random[], Random[]}, {1000}]]
Plot[0, {x, 0, 1}, Epilog -> myvertices]
