I understand you are referring to Mathematica 3 notebooks, and not the old format. If the notebook's overall Notebook[] expression has not been corrupted, try editing the notebook file on an ASCII editor and remove the notebook cache (all the lines before and after Notebook[]). This forces Mathematica to reinterpret the notebook and rebuild the file. If the Notebook[] expression has itself been corrupted, I think what can only be done is look at the expression and correct it (Mathematica indicates where the error is the first time it tries to read the file).
